The Spanish third seed was outplayed under the Rod Laver Arena roof, going down to Strycova 6-3 6-2 in the third round.
Muguruza stunned by Strycova at Aus Open
Garbine Muguruza has suffered a shock straight-sets loss to Barbora Strycova at the Australian Open on Saturday.
Muguruza, last year's Wimbledon runner-up, struggled to find her rhythm as her Czech opponent moved into the round of 16 for the first time in Melbourne.
Strycova had failed to win a set in two previous meetings with Muguruza, but she took her chances and moved the 22-year-old around from the baseline.
It continued the plethora of seeded departures in the bottom half of the draw as Muguruza joined the likes of Simona Halep, Venus Williams, Jelena Jankovic, Timea Bacsinszky and Elina Svitolina.
Muguruza won just 27 of 70 points from the baseline during her 76-minute loss.
Strycova was challenged on serve in the first set and while she failed to consolidate her opening break, there were no such issues as she took a 4-1 lead.
A break early in the second had her on her way and Muguruza was unable to find any answers, broken again in the seventh game as Strycova had no troubles closing out her win.
The 29-year-old now faces either 14th seed Victoria Azarenka or unseeded Japanese Naomi Osaka in the fourth round.
