Jordan Ayew fired Villa ahead in the 13th minute, but the hosts staged a comeback to win 3-1: Jack Gray put them level in the 35th minute.
Cameron McGeehan put Luton Town into the lead in the 60th minute, before Jores Okore connected the ball into his own net to give the league two side a 3-1 victory.
It was a big blow to Roberto Di Matteo in his quest to revive the former English Premier League out Aston Villa, as they compete in the Championship.
It is still not certain whether Jordan Ayew would remain at Aston Villa or not, since several clubs have shown the interest to secure his signature.
