Fall was included in Guy Noves' initial 31-man squad, but will now miss out with an unspecified injury.
In his place comes Montpellier team-mate O'Connor, who earns a maiden call-up to the full national team.
O'Connor, the son of a former Australian rugby league player, has also represented Les Bleus at youth level.
The 24-year-old will bid to make his first international appearance when France play Italy in their Six Nations opener at the Stade de France on February 6.
